It is then known that mixing Donkey-anti-Goat and Donkey-anti-Mouse provides a successful solution.

This antibody recognizes a protein of 75 kDa, identified as Thrombomodulin. Thrombomodulin is a transmembrane glycoprotein with natural anticoagulant properties. It is normally expressed by a restricted number of cells, such as endothelial and mesothelial cells. In addition, synovial lining and syncytio-trophoblasts of placenta also express thrombomodulin. This protein is present in almost all of benign vascular tumors and majority of malignant vascular tumors (Kaposi's sarcoma, angiosarcoma, and epithelioid hemangioendothelioma). Hence, anti-thrombomodulin serves as a sensitive marker for lymphatic endothelial cells and their tumors. This MAb recognizes a 60 kDa antigen associated with the mitochondria in human cells. It can be used to stain mitochondria in cell or tissue preparations and can be used as a mitochondrial marker in subcellular fractions. It produces a spaghetti-like pattern in normal and malignant cells. This antibody is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells, including neurons and embryonic stem cells. Immunostaining pattern with anti-mitochondrial MAb has been reported as a useful discriminatory adjunct in the complex differential diagnosis of granular renal cell tumors. This antibody recognizes a protein of 62 kDa, identified as PAX8. It is a member of the paired box (PAX) family of transcription factors. This nuclear protein is involved in thyroid follicular cell development and expression of thyroid-specific genes. Mutations in this gene have been associated with thyroid dysgenesis, thyroid follicular carcinomas, and atypical thyroid adenomas. PAX-8 is expressed in the thyroid (and associated carcinomas), non-ciliated mucosal cells of the fallopian tubes, and simple ovarian inclusion cysts, but not normal ovarian surface epithelial cells. PAX-8 is expressed in a high percentage of ovarian serous, endometrioid, and clear cell carcinomas, but only rarely in primary ovarian mucinous adenocarcinomas. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. It inhibits caspase activity either by preventing the release of cytochrome c from the mitochondria and/or by binding to the apoptosis-activating factor (APAF-1). Expression of Bcl-2 inhibits programmed cell death (apoptosis).
